Project Title:
Role of the unfolded protein response in the progression and chemo-radiosensitivity of endometrial tumors
The role of ER stress, UPR induction and BiP/Grp78 in the development and progression
of endometrial tumors, as their role in resistance to therapies, have never been
investigated. Our preliminary data indicate that, as reported in breast, prostate, gastric and
colon carcinomas, BiP/Grp78 is overexpressed also in endometrial carcinomas. Thus, ER
stress, UPR induction and BiP/Grp78 may play an important role in these tumors. This
hypothesis needs to be further confirmed and the molecular mechanisms identified. With
this background, the proposed project will pursue the following three major objectives.
_ To elucidate the role of ER stress, UPR signalling and BiP/Grp78 in endometrial
tumors development and progression both in vitro and in vivo.
_ To define the significance of ER stress, UPR signalling and BiP/Grp78 to invasion
and metastasis both in vitro and in vivo;
_ To define the significance of ER stress, UPR signalling and BiP/Grp78, to
radio/chemoresistance both in vitro and in vivo, improving therapeutic strategies.
Understanding the role of ER stress, UPR induction and BiP/Grp78 in the development
and progression of endometrial tumors, as their role in resistance to therapies, as
described in the next section, is expected to generate novel and valuable information for
identifying novel targets for prevention and therapy of endometrial cancer.
